The Intensive Programme "Diagnostic Techniques and Analysis Trends for Establishing a Health

Profile" – DTHP, has as aims and objectives the presentations by the lecturers, teachers with research activity, different fields of the modern diagnosis techniques, currently used in diseases diagnostic.

The Intensive Programme making up of an extended variety of diagnosis techniques whose complexity confers an interdisciplinary character enables the international relationships character.

According to the intensive teaching title, the lectures presented by lecturers include a wide variety of diagnosis techniques (techniques for detection and biofunctionalization of biochemical particles by biomembranes, enzymatic, immuno-enzymatic, immuno-histochemical, immunologic techniques, bioinformatics as current and future scientific field, issues from the personalized medicine that also represents perspective fields with vast implications on the patient and on the pathologies ample, intricate genetic and molecular techniques used in the diagnosis of diseases, modern methods of analyzing the mechanisms of apoptosis in pathology, technologies related to transplant and stem cells, toxicology tests applied in pathology, history of medicine in pathology).

The innovative character of the Intensive Programme is provided by the approach of the complexity of the diagnosis techniques in an extremely wide sphere of the current pathology, according to the project title.

The topics approach are according to the Intensive Programme title "Diagnostic Techniques and Analysis Trends for Establishing a Health Profile" – DTHP.

The topics are approached by lecturers from 7 partners universities ( Claude Bernard Lyon 1- University, Istanbul University, Athene University, University of Medicine Varna, Politechnique University Bucharest, Faculty of Medicine Lucian Blaga Sibiu, Transilvania University of Brasov ) and 5 countries ( France, Turkey, Greece, Bulgaria, Romania) and it creates the interdisciplinary specificity of the Intensive Programme, putting it in the European context.

The complexity of the topic by diseases diagnostic techniques, the presentation tools by CDs, as support of the teaching course with videolectures, open discussions between students (Master students and PhD students) and teachers, video teaching demonstrations and teaching practical applications used to understand each technique as well as the assessment ways by questionnaires with two types of questions with simple answer and multiple choice answer which are video presented, confer the innovative character of the Intensive Programme.

The Intensive Programme topic addressed to a complexity of fiels for research, in respect of the title, "Diagnostic Techniques and Analysis Trends for Establishing a Health Profile" – DTHP.

In this context, Enzymatic techniques is used for diagnosis reffering to the study of the enzymatic activity of a particular enzyme, given that its study is required with a view to establishing a diagnosis. Enzymology is continued in the same line with the immunoenzymatic techniques for diagnosis. Among them, ELISA technique may be mentioned, based on an antigen-antibody reaction, with applicability in detecting viral antigens, antibodies or in detecting mycotoxins, hormones, cytokines, tumour markers, drugs, as well as for instance the reaction Quantiferon TB-Gold, used in the indirect diagnosis of the infection with Mycobacterium tuberculosis.

Immunohistochemistry is the combination between immunology and histochemistry; and it resorts to monoclonal antibodies whereto enzymes are linked, coupled in their turn to portions of the antibody, with chromogenic substrates placed. The purpose of the technique is to highlight the antigenic substances in a tissue, cell or on subcellular level, of usefulness in diagnosing pathologies.

In general, immunochemical tests based on an antigen-antibody reaction are used to the purpose of determining tumour, allergologic, autoimmune and endocrine markers.

Immunochemical tests consist in marking the antigen or the antibody with a substance capable of generating a measurable signal. In this context, RIA radioimmunologic tests resort to isotope marking, EIA tests resort to enzyme marking, FIA tests resort to fluorescent substance marking and CLIA tests use, for marking,

chemiluminescence-generating molecules. ECLIA tests are also known, which resort to organic compounds for marking and detection through electrochemiluminescence.

Methods for analyzing DNA, RNA, proteins, CRP, RE, in situ hibridization inscribe within modern genetic technologies. They are combined with molecular diagnostic techniques used for diagnosis. DNA identification likewise stands for a technique used in forensic investigations. Immunological and immunoenzymatic techniques are also used in the case of the contamination with various microorganisms, CRP reaction being one of the main detection methods.

Analysis techniques upon the apoptotic mechanisms in diseases, in the context of the current pathology, are of importance in the investigations used for diagnosing cancer. Apoptotic mechanisms are controlled in two activation ways. The extrinsic way involves the tumour necrosis factor and the intrinsic way involves the mitochondria, being controlled by proteins pertaining to the family BcL-2. Apoptosis follows a sequence of steps, involving proapototic factors, caspase cascades, as well as useful pores in the transition of the mitochondria, key organelles in apoptosis.

Immunological analysis likewise represents a technique used in investigations such as stem cell transplantation, and also in numerous other types of pathologies, among whom lymphatic diseases, systemic diseases.

Some aspects in the field of personalized medicine refer to the study of the individual’s genetic profile, with a view to applying the adequate therapy, defining pharmacogenetics. Genetic predisposition tests, genomic techniques, proteomics, metabolomics, transcriptomics are used in personalized medicine. The applicability of this technique is extended into a complex range of pathologies; the neoplastic diseases and those with irradiation-risk to the patients through exposure to the imaging-investigation equipment being of interest.

A category of modern techniques currently used for diagnosis is represented by the electron microscopy studies, which allow ultrastructural investigations upon the fragments of tissue and/or organ, to the purpose of establishing the modifications that appear on cellular and subcellular level, cell membrane, cell organelles, nucleus. The ultrastructural modifications of the cellular and subcellular components occur in variegated pathological affections, with reference to all organs. Among the electron microscopy techniques used in studying current pathology, TEM, SEM and LSEM may be mentioned.

Since the pathology of our days is complex, the investigation techniques used for diagnosis refer to pulmonary, cardiovascular, gastrointestinal, neurological, neurodegenerative and psychiatric pathology, to the study of cancer, the mechanisms involved in organ transplantation, systemic diseases, the applicability in medicine of the contamination with microorganisms; therefore the multidisciplinary character of IP can be noticed.

To the purpose of good Intensive Programme proceedings, it is envisaged to prepare the didactic material for the Master students and PhD students, on electronic course support, in the form of CDs, as video projections, together with questionnaire videopresented, using questions by topics, composed with simple or multiple-choice answers, in order to assess the knowledge retention by students. These are preceded by informal discussions on each topic, unfolding between teachers (lecturers) and students (Master students and PhD students) and are followed, after the application of the assessment survey, by practical teaching demonstrations and video demonstrations, with a view to retaining the taught technique and to preparing the Master students and PhD students’ final assessment for obtaining Diploma Supplement and ECTS.
